摘要 为保存野生资源,满足栽培对种苗的需要,以牛蒡嫩茎为材料,应用组织培养的方法,进行了嫩茎愈伤组织诱导与分化、分化芽生根、试管苗的生根继代及移栽和定植的研究。结果表明:MS+6-BA0.3mg.L-1+2,4-D2.0mg.L-1是嫩茎愈伤组织诱导培养和继代增殖培养的理想培养基;MS+6-BA1.5mg.L-1+NAA0.1mg.L-1+AgNO31.2mg.L-1是愈伤组织分化培养的理想培养基;White+NAA0.1mg.L-1+IAA0.2mg.L-1是不定芽生根培养、试管苗生根继代培养及快速繁殖的理想培养基。试管苗易移栽和定植易成活,且定植的试管苗保持了野生牛蒡的所有生物性状。 In order to preserve the wild resources and meet the needs of germchits for planting,with the method of tissue culture,tender stems of Arctium lappa were used as material to do the research on callus inducement and differentiation,rooting of adventitious buds and rooting multiplication of tube seedlings,transplantation of tube seedlings.The results show that the ideal medium for callus induction and multiplication of tender stems was MS +6-BA0.3mg·L-1 + 2,4-D2.0mg·L-1;MS+6-BA1.5mg·L-1+NAA0.1mg·L-1+AgNO31.2mg·L-1was the best mediums for callus differentiation;the medium White+NAA0.1mg·L-1+ IAA0.2mg·L-1 was suitable for rooting of adventitious buds,tube seedlings and rapid propagation.The test tube seedlings transplanted were easy to survival and maintained all biological characteristics of the wild Arctium lappa.
