摘要
目的研究TGF-β1修饰的供者脾细胞特异性输注对大鼠移植肝脏耐受的诱导及效果。方法每组选10只雄性SD大鼠为供体,10只雄性Wistar大鼠为受体建立大鼠原位肝脏移植模型,于移植前7天分别经尾静脉输注TGF-β1基因修饰的脾细胞(5×106/mL)1mL、脾细胞(5×106/mL)1 mL、空质粒1 mL、生理盐水1 mL。观察TGF-β1修饰的脾细胞输注组、脾细胞输注组、空质粒输注组、肝移植组存活天数和病理改变及凋亡情况。结果 TGF-β1修饰的脾细胞特异性输注可明显延长移植肝脏存活时间(36.4±4.16)d,与脾细胞输注组(20.0±4.00)d、空质粒输注(7.8±0.84)d、肝移植组(8.4±1.4)d比较,差异有统计学意义(P<0.05);而且可明显减轻移植肝脏的病理损伤。结论输注TGF-β1基因修饰的供体脾细胞可诱导大鼠移植肝脏的耐受产生。
Objective To study whether TGF-β1 gene-modifide donor spleen cell transfuntion could induce immune tolerance of liver transplatation.Methods In each group,selected 10 male Sprague-Dawely rats as donor and 10 male Wistar rats were established as a receptor for orthotopic liver transplantation.7 days before transplantation,TGF-β1 gene-modified spleen cells(5×106/mL) 1 mL,spleen cells(5×106/mL) 1 mL,empty vector 1 mL,normal saline 1 mL were infused via the tail vein,respectively.Survival time,the pathological changes and apoptosis were observed in each group.Results TGF-β1-modified spleen cells transfusion could significantly prolong the survival time of orthotopic liver transplantation(36.4±4.16)d,compared with spleen cell infusion group(20.0±4.00) d,empty plasmids infusion(7.8±0.84)d,liver transplantation group(8.4±1.4)d,there was significantly different among four groups(P0.05);and could significantly reduce the pathological damage of the transplanted liver.Conclusion The infusion of TGF-β1 gene modified donor spleen cells can induce the production of rat liver graft immune tolerance.
