摘要
目的探讨蟾蜍灵(bufalin)对多发性骨髓瘤(multiple myeloma,MM)的白细胞介素-6(interleukin-6I,L-6)表达的影响,以期证实蟾蜍灵抗MM的分子生物学机制。方法人MM细胞系U266与不同浓度的蟾蜍灵作用,逆转录聚合酶链反应(RT-PCR)检测IL-6基因表达。结果不同浓度的蟾蜍灵作用U266细胞48h后RT-PCR检测结果表明:U266细胞的IL-6基因表达与对照相比均明显降低;差异有显著性意义(P<0.01)。结论蟾蜍灵能抑制U266细胞的IL-6基因表达。提示降低IL-6的表达是蟾蜍灵抗MM的机制之一。
[ Objective ] To investigate the mechanism ofbufalin on the inhibition of human muhiple myeloma (MM) cells. [ Methods ] order to evaluate the mechanismof bufalinon the inhibition of MM. Expression of IL -6 were evaluate by RT - PCR methods after treatment with different concentrations ofbufalin. [ Results ] The expression of IL -6 gene was significantly decreased after treatment withbufalin compared withouthufalin. [ Conclusion ] U266 cells, bufalin significantly inhibit the expression of IL - 6 which seems to be one of mechanisms of anti - MM effects ofbufalin.
