摘要
目的探讨αSMA、MMP-13及VEGF在应用枸杞多糖治疗动脉粥样硬化家兔动脉中的表达及临床意义。方法将24只雄性新西兰白兔随机分成3组:空白对照组、模型组、预防组,每组8只。空白对照组喂饲普通饲料,模型组喂饲含2%胆固醇的饲料,预防组喂饲含2%胆固醇的饲料,并在每天早晨饲料中按体重加入枸杞多糖(LBP)20mg/kg。第16周末空气栓塞处死所有动物并观察主动脉形态学变化及应用免疫组织化学染色进行指标的检测及分析。结果与空白对照组相比,模型组全部形成动脉粥样硬化模型,预防组也形成了动脉粥样硬化,但是斑块厚度明显低于模型组。通过免疫组织化学染色发现:预防组与模型组比较,主动脉斑块厚度和斑块内平滑肌细胞明显减少;血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达降低,预防组与模型组相比,VEGF的阳性表达率显著减小(P<0.05);MMP-13(Matrix metalloproteinase 13)的表达降低,预防组与模型组相比,MMP-13的阳性表达率显著减小(P<0.05)。结论 1.口服枸杞多糖能明显抑制高脂血症所诱发的VSMC增殖,促进已经增殖的VSMC凋亡;2.枸杞多糖能够降低动脉斑块内MMP-13和VEGF的表达,从而抑制动脉粥样硬化的发生与发展;3.口服枸杞多糖能有效预防高脂饲料诱发的家兔主动脉AS斑块的形成。

Through the immunohistochemical found: prevent group group compared with model, aortic plaques within the thickness and plaques smooth muscle cell significantly reduced; Vascular endothelial growth factor (vascular endothelial growth factor, VEGF) expression is reduced, prevent group compared with model group, VEGF positive expression rate reduced significantly (P 〈 0.05 ) ; MMP - 13 (Matrix metalloproteinase 13) expression is re- duced, prevent group compared with model group, MMP- 13 positive expression rate reduced significantly (P 〈 0. 05). [ Conclusion ] 1. Oral LBP significantly suppress hyperlipidemia induced by VSMC proliferation, promote the proliferation of VSMC has apoptosis ;2. LBP can reduce artery plaque inside MMP - 13 and the expression of VEGF, thus inhibit atherosclerosis occurrence and development;3. Oral LBP can effectively prevent the high fat feed induced rabbit aorta AS patch formation.
