实时荧光定量PCR技术在副溶血性弧菌快速检测中的应用 被引量:4

Application of real-time quantitative PCR in the rapid detection of Vibrio parahaemolyticus in food and food poisoning samples

摘要 目的:建立副溶血性弧菌荧光定量PCR检测方法,应用于食品、食物中毒样本的快速检测。方法:食品样本先选择性增菌,取上层液提取DNA,粪样直接提取DNA,用实时荧光定量PCR方法进行检测,并与常规培养法进行比较。结果:用实时荧光定量PCR检测食品54份,食物中毒粪样12份,检测阳性食品21份、粪样9份,检测结果与培养法比较,阳性检出率分别为食品38.9%、31.5%;粪样均为75.0%。结论:副溶血性弧菌的实时荧光定量PCR检测方法具有快速、灵敏的优点。 Objective:The Vibrio parahaemolyticus quantitative PCR method was established for rapid detection of food and food poisoning samples.Methods:Selective enrichment of food samples was done at first,then DNA was extracted from upper liquid and stool samples,finally real-time quantitative PCR was used for detection and compared to the routine culture method.Results:54 food samples and 12 stool samples of food poisoning were detected by real-time PCR.The results showed 12 food samples and 9 stool samples were positive.Compared to the culture method,the positive rates of food samples were 38.9% for PCR and 31.5% for culture method,while that of stool samples were both 75.0%.Conclusion:Real-time quantitative PCR was an ideal method for rapid Vibrio parahaemolyticus detection with high accuracy and sensitivity.
