目的:研究MicroRNA-143在膀胱癌中通过抑制EMT机制,降低肿瘤浸润、转移的风险,从而延缓膀胱癌演进。材料和方法:分别测定不同分期分级的膀胱癌病例标本及膀胱癌细胞株中MicroRNA-143、c-myc及E-Cadherin的表达,分析它们之间统计学意义;转染MicroRNA-143及c-myc的siRNA至浸润性膀胱癌细胞株中,测定E-Cadherin表达,通过侵袭及迁移等生物学行为测定EMT机制。结果:不论病例标本还是细胞株,恶性程度高的膀胱癌中MicroRNA-143、E-Cadherin表达均低于浅表性膀胱癌,而c-myc则反之;转染MicroRNA-143及c-myc的siRNA至浸润性膀胱癌细胞株中后,E-Cadherin表达增高,细胞株的侵袭及迁移能力降低,EMT受抑制。结论:MicroRNA-143通过下调其靶基因c-myc,致使E-Cadherin表达增加,抑制EMT机制,延缓肿瘤演进。
Objective To study the role of MicroRNA-143, by inhibiting EMT mechanism, in reduction of tumor invasion and metastasis risk and thus in delaying the progression of bladder cancer. Materials and Methods MicroRNA-143, c-myc and E-Cadherin expression at different stage and grade of bladder cancer cases were measured in both specimen and bladder cancer cell lines and statistical significance between them was analyzed. siRNA of MicroRNA-143 and c-myc was transfected to invasive bladder cancer cell lines and E-Cadherin expression was evaluated. EMT mechanism of bladder cancer cells was determined through their biological behavior like invasion and migration,Results MicroRNA-143 and E-Cadherin expression were found to be lower in both specimen and cell lines of highly malignant bladder cancer cases when compared to that of superficial bladder cancer cases, while expression of c-myc levels was found to be just opposite. After transfecting siRNA of MicroRNA-143 and c-myc to invasive bladder cancer cell lines, E-Cadherin exhibited increased expression while invasion and migration ability of tumor cells was reduced and their EMT mechanism was inhibited..Conclusions Through down-regulation of target gene c-myc and resulting increased expression of E-Cadherin, MicroRNA-143 inhibits tumor cell EMT mechanism to delay progression of bladder cancer.
