摘要 目的对地榆炭配方颗粒进行薄层色谱鉴别及HPLC含量测定研究,以客观评价地榆炭配方颗粒的质量。方法薄层色谱鉴别以没食子酸对照品为对照,以甲苯(用水饱和)-乙酸乙酯-甲酸(6:3:1)为展开剂,喷以1%三氯化铁乙醇溶液,日光下检视;含量测定采用Ultimate XB-C18色谱柱(4.6×200mm,5μm),流动性为甲醇-0.05%磷酸溶液(5:95),流速:1.0 mL/min,检测波长:272 nm。结果地榆炭配方颗粒在对照品相同位置上显相同颜色的斑点;含量测定中没食子酸在5.115-102.3μg呈良好的线性关系,样品中没食子酸峰分离度良好。结论首次建立了地榆炭配方颗粒中没食子酸的薄层色谱鉴别方法,并进行了没食子酸的含量测定,提供了控制地榆炭配方颗粒质量的科学依据。 Objective To evaluate the quality of Burnet carbon formula particles by TLC and HPLC. Methods Gallic acid was identified by TLC with toluene(saturated with water)-ethyl acetate-formic acid(6:3:1) as solvent,1% solution of ferric chloride in ethanol as reagent;An HPLC method was performed on Ultimate XB-C18 column(4.6 × 200 mm,5 μm) with methanol-0.05% phosphoric acid(5:95) as mobile phase.The flow rate was1.0mL/min,detection wavelength was set at 272 nm..Results The Burnet carbon formula particles had the same color spot with the reference substance at the same position;The calibration curve of gallic acid was linear in the range of 5.115-102.3 μg,and the peak showed a good separation.Conclusion We established a TLC distinguish method and an HPLC content determination method of gallic acid in the Burnet carbon formula particles for the first time,and provided the scientific basis for the quality control of Burnet carbon formula particles.
