摘要
病菌和外来异物入侵时,对虾血细胞在吞噬过程中存在利用活性氧杀菌防御机制。对中国对虾血细胞体外吞噬现象进行观察,利用化学发光法和NBT法检测血细胞体外吞噬过程中产生的活性氧,以研究不同浓度壳寡糖对活性氧作用的影响。结果发现,对虾的血细胞在体外具有一定的吞噬能力,吞噬率为42.7%±0.80%;吞噬指数为1.34±0.11;随着加入壳寡糖浓度的提高,化学发光法和NBT法检测数值具有相同的趋势,其中1.0mg/L,5.0mg/L以及10mg/L壳寡糖对活性氧的产生具有明显的增强效果(P<0.05)。结果表明,采用的化学发光法与NBT法可以检测出血细胞吞噬过程中产生的活性氧,并证实壳寡糖对对虾的血细胞产生的活性氧有不同程度的诱导增强作用。
To study the influnce of oligochitosan on active oxygen during phagocytosis,the phagocytosis of Penaeus chinensis hemocytes were observed in vitro;the active oxygen was detected by Chemiluminescence immunoassay(CLIA) and Nitroblue tetrazolium(NBT) reduction assay.The results indicate that the hemocytes have the phagocytic activity in vitro,percent phagocytosis and phagocytic index are 42.7%±0.80 and 1.34±0.11 respectively;With oligosaccharides concentration increased,the changs of chemiluminescence and NBT assay value have the same trend;The 1.0 mg/L,5.0 mg/L and 10 mg/L oligosaccharides can enhance the generation of reactive oxygen significantly(P0.05).The results show the active oxygen can be meatured by chemiluminescence and NBT assay which confirm that oligosaccharides can induce the changes of reactive oxygen during phagocytosis Penaeus chinensis hemocytes.
