摘要
目的:研究玉郎伞(YLS)皂苷对大鼠离体心脏缺血再灌注损伤(MI/RI)的保护作用及其作用机制。方法:40只Wistar大鼠随机分为5组(每组8只):正常对照组,缺血-再灌注组,YLS皂苷低剂量组(2×10-2g·L-1),YLS皂苷高剂量组(4×10-2g·L-1),阳性对照维拉帕米组(5×10-7mol·L-1)。离体大鼠心脏采用Langendorff法灌流,停灌30 min再灌30 min造成心肌缺血-再灌注损伤模型。左心室插入水囊导管,记录YLS皂苷对血流动力学指标的影响,测定冠脉流量(CF)和冠脉流出液中肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)、乳酸脱氢酶同工酶-1(LDH-1)活性以及心肌组织超氧化物歧化酶(SOD)和丙二醛(MDA)含量。结果:YLS皂苷高剂量可显著改善缺血-再灌注所致的心功能损伤,减少CK,CK-MB,LDH,LDH-1的释放和心肌组织MDA的产生,增加SOD的活性(与模型组相比,P<0.05或P<0.01)。结论:YLS皂苷对心肌缺血-再灌注损伤具有保护作用,其机制可能与清除氧自由基、减少脂质过氧化反应有关。
Objective: To study the effect and mechanism of saponin of Yulangsan(SYLS) on myocardial ischemia reperfusion injury in isolated rat hearts.Method:Fourteen Wistar rats were randomly divided into five groups: normal group,ischemia reperfusion group,verapamil group(5×10-7 mol·L-1),and SYLS groups(2×10-2,4×10-2 g·L-1).Langendorff-perfusion isolated rat hearts were prepared.Myocardial ischemia reperfusion injury was induced by 30 minutes global ischemia followed by 30 minutes reperfusion.The effect of SYLS on cardiac hemodynamics was observed by MPA-CFS.The activities of creatine kinase(CK),creatine kinase isoenzyme MB(CK-MB),lactate dehydrogenase(LDH),lactate dehydrogenase isozyme-1(LDH-1) in the effluent from coronary artery were measured to evaluate the injury of myocardial cells and the protective effect of SYLS.Meanwhile,the activity of superoxide dismutase(SOD) and the contents of malondialdehyde(MDA) in myocardial tissues were determined.Result: Compared with the I/R group,high dosages of SYLS could recover cardiac function,and reduce the releases of CK,CK-MB,LDH and LDH-1 from I/R rat hearts.It could increase the activity of SOD,and decrease the MDA product(P0.05 or P0.01).Conclusion:SYLS may offer myocardial protective effects against ischemia reperfusion injury,which may be related to its effects of scavenging oxygen free radical,reducing lipid peroxidation.
