新疆哈族和汉族食管癌中Toll样受体的表达及其与肿瘤细胞浸润和转移的关系 被引量:2

Expression of Toll-like receptor in esophageal cancers and their correlation with local invasion and lymphnode metastasis in Xinjiang Kazak and Han

摘要 目的探讨Toll样受体TLR 4、7、9 mRNA和蛋白在新疆不同民族食管癌组织中的表达及其与肿瘤浸润和转移之间的关系。方法应用实时荧光定量PCR(qRT-PCR)法检测40例食管癌及相应癌旁组织中TLR4、7、9 mRNA的表达。采用免疫组化SP法检测90例食管癌及相应癌旁组织中TLR4、7、9蛋白的表达。结果 TLR7、9 mRNA在食管癌中的表达量明显高于癌旁组织(P<0.05);TLR4 mRNA在食管癌中的表达量与癌旁组织中的表达,差异无统计学意义(P>0.05)。TLR4、7、9 mRNA在汉族食管癌患者组织中的表达均高于新疆哈萨克族(哈族)(P<0.05)。TLR4、7、9蛋白在食管癌组织中的表达明显高于癌旁组织(P<0.05)。TLR4表达与肿瘤淋巴结转移密切相关(P<0.05);TLR7表达与肿瘤浸润深度相关(P<0.05);TLR4、7、9蛋白表达与肿瘤的分化程度密切相关(P<0.05)。TLR4蛋白在食管癌组织间质炎细胞中高表达与肿瘤的浸润和转移有关,而TLR9蛋白在食管癌组织间质成纤维样细胞中高表达与肿瘤的低浸润和低转移密切相关(P<0.005)。结论TLR4、7、9在食管癌中的高表达与食管癌生物学行为密切相关,提示该基因在食管癌的发生、发展、浸润及转移中起一定作用,其族群差异可能反应不同族群的遗传背景及肿瘤的发病机制。 Purpose To evaluate the relationship between the expression of Toll-like receptors 4,7 and 9 in Xinjiang Kazak and Han patients with esophageal cancers(EC) and that with local invasion and lymphnode metastasis.Methods The mRNA of TLR4,TLR7 and TLR9 in 40 cases of esophageal cancer and its adjacent tissues were detected with real-time fluorogentic quantitative PCR(qRT-PCR),and their protein detected with IHC method from 90 cases of esophageal cancer and its adjacent tissues.Results The quantification of TLR7 and TLR9 mRNA in EC tissues were(0.72±0.59) and(0.07±0.06),respectively,which were significantly higher than that in adjacent tissues(0.45±0.36) and(0.01±0.01),respectively(P0.05),while no significant changes of TLR4 mRNA in EC and its adjacent tissues(P0.05).The quantification of TLR4,7,9 mRNA in Han EC was significantly higher than that in Kazak EC(P0.05).The protein expression of TLR4,TLR7 and TLR9 in EC were 66/90(73.2%),68/90(77.6%) and 67/90(75.4%),respectively,which were significantly higher than that in adjacent tissues(18/90(20%),17/90(18.9%) and 30/90(33.3%,P0.05).There was a close relationship between the protein expression of TLR4 and lymph node metastasis(P0.05).TLR7 overexpressed in EC was remarkably associated with tumor invasion(P0.05).TLR4,TLR7 and TLR9 overexpressed in esophageal cancer was remarkably related with tumor differentiation degree(P0.05).The higher protein expression of TLR4 in EC interstitial cells was correlated with invasion and lymph node metastasis.The higher expression of TLR9 in fibroblast cells of EC interstitial tissue was remarkably related with decrease of invasion and lymph node metastasis(P0.005).Conclusions Higher expression of TLR4,TLR7 and TLR9 in EC is significantly associated with EC carcinogenesis,invasion and metastasis.These differences between ethnic groups may reflect the different genetic background and the tumor pathogenesis.
