过氧化物酶体增殖物激活受体γC161→T多态性对学龄儿童肥胖及血脂的影响

EFFECT OF PEROXISOME PROLIFERATORS ACTIVATED RECEPTORS GAMMA C161→T POLYMORPHISM ON SCHOOL CHILDREN OBESITY AND SERUM LIPID PROFILES

摘要 目的研究过氧化物酶体增殖物激活受体γC161→T多态性在学龄儿童中的分布及其与学龄儿童肥胖和血脂、血糖等血生化指标的关系。方法按照整群分层抽样方法,从上海某区6所学校2-5年级抽取1173名小学生作为研究对象,分别进行体格检查和空腹采血,测定血清中血脂、血糖、胰岛素,血凝块中提取基因组DNA,采用聚合酶链反应和限制性片段长度多态性方法(PCR-RFLP)检测研究对象PPARy C161→T的基因型。结果 (1)男生超重肥胖率25.4%,女生超重肥胖率13.7%;(2)调查人群中,CC、CT、TT三种基因型分别占52.5%,41.2%,6.3%,三种基因型分布和等位基因携带率在正常体重、超重、肥胖中无差别,在性别分布中也无不同,但是T突变基因是女性学龄儿童超重肥胖的危险因素;(3)T突变基因携带者血清中TCH含量高,女性学龄儿童T突变基因携带者血清TCH、TG浓度高,正常体重儿童T突变基因携带者TCH含量高,超重肥胖儿童TG含量高。结论上海市学龄儿童肥胖超重患病率比2005年显著增加,PPARy C161→T突变对女性儿童是肥胖和血脂增加的危险因素。 Objective To study the genotype distribution of peroxisome proliferators activated receptors gamma(PPAR(?))C161→T gene in school children and its possible association with overweight/obese and serum glucose,lipidlevels.Method A total of 1173 primary school students aged 7 to 12 in a district of Shanghai were cluster sampled,andtheir physical examination,the levels of serum lipids profile,glucose,and insulin were measured.The gene polymorphism of PPARγC161→T variants was decided via methods of PCR-RFLP.Results(1)The prevalence of overweight/obese ofboys and girls was 25.4%and 13.7%respectively.(2)The frequencies of CC,CT,TT in study participants were 52.5%,41.2%,and 6.3%respectively.There was no significant difference of the genotype and allele distribution in PPARγC161→T in different weight groups and between boys and girls.And T allele was associated with higher prevalence ofoverweight/obese in girls.(3)The level of serum TCH of participants with T allele was higher compared to those with CCgenotype.And the concentrations of serum TCH,TG of girls,TCH of normal weight groups,and TG of overweight/obesewith T allele were higher than the level of those with CC genotype.Conclusion The prevalence of overweight/obese inShanghai was higher as compared with that in 2005,and T allele was the risk factor of obesity and hyperlipemia in girls.[ACTA NUTRIMENTA SINICA,2012,34(2):123-127]
