摘要
研究整合素阻断多肽HM-3体外对血管生成的作用。应用鸡胚尿囊膜实验、大鼠动脉环实验及Matrigel实验考察整合素阻断多肽HM-3抗血管生成作用。结果:鸡胚尿囊膜实验和大鼠动脉环实验表明HM-3能够抑制体外血管生成,多肽HM-3 0.05μg对鸡胚尿囊膜新生血管生成的抑制作用最强,多肽HM-3 8μg/mL对大鼠动脉环微血管生成抑制作用最强。Matrigel实验表明HM-3 7~8μg/mL对内皮细胞生长因子添加剂(ECGS)诱导的HUVEC细胞体外分化成管状结构抑制作用最强。整合素阻断多肽HM-3具有显著的体外抗血管生成的作用。
To investigate the inhibitory effect of HM-3 on angiogenesis in vitro.HM-3 antiangiogenesis effect was evaluated by using chick chorioallantonic membrane,rats aortic rings and Matrigel assay.Chick chorioallantoic membrane and rats aortic rings tests demonstrated that HM-3 effectively inhibited angiogenesis in vitro,the peptide HM-3 at the dose of 0.05 μg revealed the best inhibitory effect on chick chorioallantoic membrane.The most obvious effect on angiogenesis inhibition was observed at dose of 8 μg/mL on rats aortic rings.The most obvious blocking effect on the differentiation of HUVECs into tube-like structures was observed in vitro when the concentration of HM-3 was 7-8 μg/mL.Therefore HM-3 distinguishedly inhibited angiagenesis formed in vitro.
