十枣汤对荷瘤小鼠p53突变及内皮素表达的影响

The Impacts of Shizao Decoction to p53 Mutation and Expression of Endothelin in Tumor-bearing Mice

目的观察十枣汤对荷H22腹水瘤种小鼠的抗肿瘤作用。方法小鼠随机分为空白对照组、荷瘤模型组、环磷酰胺(CTX)阳性对照组和十枣汤高、中剂量组。除空白对照组外,其余4组小鼠均以皮下注射瘤细胞悬液0.2 mL造模。于造模第2天,分别灌胃相应药物或生理盐水,或注射CTX。采用免疫组化方法检测瘤体内突变的p53蛋白;放射免疫法测定小鼠血浆内皮素(ET)水平。结果各用药组突变型p53表达水平明显低于荷瘤模型组(P<0.01),而3组间比较差异无显著性意义(P>0.05);模型组小鼠血浆ET水平明显高于空白对照组(P<0.01),各用药组ET水平均有不同程度下降(P<0.05,P<0.01)。结论十枣汤可以降低突变型p53蛋白表达,降低血浆ET水平并减少肿瘤的血供,这可能是其发挥抗肿瘤作用的机制之一。

Objective Investigate the anti-tumor effect and the possible treatment mechanism of shizao decoction to mice bearing H22 with ascites tumor.Methods 75 healthy mice were randomly divided into 5 groups,which were the blank control group,tumor model group,positive control group and high and middle dose of Shizao Decoction group.Except the blank control group,the other mice of four groups were given subcutaneous injection of tumor cell suspension,0.2 mL/only to establish model.The next day after modeling,the blank control group and tumor model group were fed with normal saline（NS）.The mice of high and middle dose group were given 100% and 50% concentration of Shizao Decoction to feed for 10 days,two times a day.The mice of positive control group were given CTX 10 mL/kg intraperitoneal injection for 5 times,Once every other day.Used immunohistochemical method to detect mutant p53 protein in vivo tumor and measured the levels of mouse plasma endothelin（ET）by radioimmunoassay.Results The expressing mutant p53 levels of tumor model group were significantly higher than the other three groups（P0.01）,and three treatment groups have different degree of decline,but the difference among these three groups was not significant（P0.05）.The plasma level of ET in tumor model group was significantly higher than that in blank control group（P0.01）.All the dose groups,ET levels were decreased in different degrees,especially the high dose group that was close to the blank control group.Conclution Shizao Decoction can reduce the expression of mutant p53 protein,plasma ET levels and the tumor＇s blood supply.These may be one of the anti-tumor effect mechanisms of shizao decoction.
