摘要
目的分离筛选出白蔹抗肿瘤的活性部位,并初步探讨其作用机制。方法采用溶剂法和硅胶柱层析法对白蔹醇提物进行不同部位分离,运用四甲基偶氮唑盐(MTT)法对各部位进行体外抗肿瘤活性测定,Hochest/PI双染色法观察白蔹活性提取物抑制细胞增殖的作用。结果在25-200 mg/L范围内,乙醚和乙酸乙酯部位及从乙酸乙酯部位分离得到的P2、P3组分对人肝癌HepG2细胞的增殖有较强的抑制作用,且具有明显的浓度依赖性。结论乙醚和乙酸乙酯部位是白蔹抗肿瘤的活性部位,并能够引起HepG2细胞凋亡和死亡。
Objective To separate and select anti-tumor activity site of Ampelopsis japonica Mak,and explore its machine processed.Methods The anti-tumor activity of the different extracts and fractions were determined by MTT assay.The effects of active site on the apoptosis of HepG2 cells were studied by Hochest/PI staining.Results In the range of 25-200 mg/L,all of the ethylether and acetate position,fractions P2 and P3,strongly inhibited cell proliferation of HepG2,and the suppression ratio was raised with increasing concentrations of drug.Conclusion The ethylether and acetate positions are the effective anti-tumor parts from Ampelopsis japonica Mak,Which can kill and induce apoptosis of HepG2 cells.
