摘要
目的探讨中医轻度"寒淫致病"的差异基因表达及相关信号通路的调节机制。方法受试者暴露在4~12℃的环境中90 min,提取其冷暴露前、后的静脉血,用全基因组芯片,采用SAM统计系统分析基因芯片,并进行RT-PCR验证。结果获得具有生物学统计意义的41条上调基因以及糖酵解/糖异生过程、三羧酸循环和氧化磷酸化代谢途径,经过验证发现与轻度"寒淫致病"密切相关的能量代谢基因ATP6V0D1基因以及与此相关的氧化磷酸化代谢通路。结论轻度的寒淫刺激机体主要是引起机体能量代谢活跃,该基因在机体能量调节方面发挥着重要的作用,有力地补充了病理状态下的中医寒证领域的研究,为中医寒淫研究的进一步深入起到了有力的提示性作用。
Objective It is to explore Chinese medicine that mild "cold pathogen induces disease" in human models,including differential gene expression and regulation of signaling pathways related.Methods The subjects exposed in 12 to 4 ℃ environment for 90 minutes,and their vein blood was extracted before and after the cold stimulation to detect blood sample with Whole-genome gene chip,using SAM statistical analysis and the RT-PCR validation.Results 41 up-regulated genes which had biological statistics significance,some changes in glycolysis/gluconeogenesis process,the citric acid cycle and oxidative phosphorylation pathway were found.After RT-PCR validation,the results prompted that the course of "cold cause disease" was closely related to the energy metabolism genes,ATP6V0D1 gene,which was associated with oxidative phosphorylation pathway.Conclusion Mild cold pathogen mainly stimulate the body's energy metabolism activity,the related genes play an important role in the traditional Chinese medicine under the cold syndrome research.In the field,the study of traditional Chinese medicine played a powerful suggestive effect.
