摘要 以自由细胞的酶作为催化剂,研究了菌体稀释倍数、丙烯腈浓度、温度、pH值、菌悬液用量和丙烯酰胺浓度对腈水合酶活性的影响.结果表明,温度、丙烯腈浓度和丙烯酰胺浓度是影响酶活性的主要因素.反应温度在28℃时酶活性达到最大为4091U/mL菌液,丙烯腈浓度采用40g/L.当反应体系中丙烯酰胺浓度为40%时,其酶活只有丙烯酰胺浓度为5%时酶活的39.2%.对不同反应温度下酶活的研究,得出腈水合酶催化反应的活化能为40.291kJ·mol-1. Taking the enzyme of free cell as the catalyst, the effects of dilution factor, concentration of acrylonitrile, temperature, pH value, concentration of ceils and concentration of acrylmide on the activi- ty of nitrile hydratase were studied. The result shows that the temperature, the concentration of acrylo- nitrile and the concentration of acrylamide are the most important among these factors. The activity of nitrile hydratase is 4 901U/mL(broth) at the reaction temperature 28℃ the concentration of acryloni- trile is 40g/L;the activity of nitrile hydratase in the solution of 40 % acrylamide is just 40% of that in so- lution of 5 % acrylamide. It is found that the activation energy of the nitrile hydratase is 40. 291kj.mol-1 through the research at the different reaction temperature.
