摘要
细胞穿透肽(cell-penetrating peptides,CPP)是一类能够携带外源物质进入细胞的多肽,因其结构和功能的独特性和实用性,近20年里在生物医学领域得到了长足的发展。目前CPP向体内和体外输送物质的方法逐渐成熟。在基础研究领域,已成为研究细胞内分子相互作用的有用工具;在应用方面,CPP药物已进入临床前、临床Ⅰ和Ⅱ期试验,在肿瘤、中枢神经系统疾病和心血管疾病的治疗中显示出明显优势。本文对CPP的发展历程、分类、入胞机制和应用等做一综述。
Cell-penetrating peptides(CPP) are a type of polypeptides that can serve as vehicles for the delivery of exogenous molecules into cells.Because of their unique structure and function,CPP research in biomedical field has been develop ing rapidly during the last 20 years.Nowadays,the methods of CPP-mediated delivery ex vivo and in vivo have gradually become mature.In the field of basic medical sciences,CPP provide a promising tool for the study of molecular interaction in cells.In the field of application study,several CPP drugs have been introduced to preclinical study,phase Ⅰ and Ⅱ clinical trials,and have shown some advantages in the treatment of some refractory diseases,such as cancers,central nervous system diseases,and cardiovascular diseases.In this article,the development process,examples of recent research,idea of classification,uptake mechanisms,and the therapeutic potential of CPP are reviewed.
