通管方对输卵管炎性阻塞性不孕大鼠模型血清IL-1、TNF-α含量的影响

Effect of "Tongguan Decoction" on IL-1 and TNF-α in rat models with tubal infertility

目的观察通管方对输卵管炎性阻塞性不孕大鼠输卵管组织形态学及血清中白介素-1(IL-1)、肿瘤坏死因子-α(TNF-α)含量的影响。方法以Wistar雌性大鼠为受试对象,采用混合菌接种法注入输卵管复制输卵管炎性阻塞性不孕模型。将180只雌性大鼠按随机排列表法抽样30只为正常组,余150只按上述方法造模,造模20天后(除去因造模所致死亡的25只)剩余大鼠再按随机排列表法分为模型组24只、血府逐瘀片组25只及通管方高、中、低剂量组,每组24只。每组采用相应的干预措施,分别于第15天、30天取材,然后采用HE染色观察输卵管组织形态学变化,采用ELISA法检测血清中IL-1和TNF-α含量。结果①用混合菌接种法注入输卵管复制大鼠炎性阻塞性不孕模型成功。②与模型组比较,通管方高、中、低组和血府逐瘀片组能不同程度地降低模型大鼠血清中IL-1、TNF-α的含量,提高受孕率,差异有统计学意义(P<0.05,P<0.01);通管方高、低剂量组与血府逐瘀片组比较,IL-1、TNF-α水平差异有统计学意义(P<0.05,P<0.01)。③治疗15天与30天比较,血府逐瘀片组和通管方各剂量组IL-1差异有统计学意义(P<0.01),通管方各剂量组TNF-α差异有统计学意义(P<0.01)。结论通管方可改善输卵管炎性阻塞性不孕大鼠输卵管的结构和功能,对血清IL-1、TNF-α的含量有调节作用,同时通管方对血清中IL-1、TNF-α的改善作用,治疗30天后的作用较治疗15天更好。

Objective To explore the effect of ＂Tongguan Decoction＂ on IL-1 and TNF-α in rats with tubal infertility and discuss the mechanism of ＂Tongguan Decoction＂.Methods Duplicated the model by mixed bacteria inoculation method.Thirty rats in 180 female rates were randomized into normal group,and the rest of 150 rats were used to establish the models.After 20 days,the 150 rats were randomized into model group（24 rats）,＂Xufu Zhuyu Tablet＂ group（25 rats） and ＂Tongguan Decoction＂（24 rats in high,medium and low dose）.Each group was divided into 2 periods,took the samples at 15 and 30 days.The IL-1 and TNF-α were detected by ELISA method,and the histomorphological changes of tubal were observed by hematoxylin-eosin staining.Results We duplicated the model by mixed bacteria inoculation method successfully.Compared with model group,the IL-1 and TNF-α were lower,while pregnancy rate was increased in ＂Xufu Zhuyu Tablet＂ group and ＂Tongguan Decoction＂ groups,with significant difference（P0.05,P0.01）;there was significant difference between ＂Tongguan Decoction＂ groups（high,low dose） and ＂Xufu Zhuyu Tablet＂ group（P0.05,P0.01）.The IL-1 in control group and ＂Tongguan Decoction＂ groups after treatment for 15 days were statistically different from 30 days（P0.01）;the difference of TNF-α among ＂Tongguan Decoction＂ groups was significant（P0.01）.Conclusion ＂Tongguan Decoction＂ can improve the structure and function of fallopian tube in rat with tubal infertility,and cure the tubal infertility in rats by regulating IL-1 and TNF-α.＂Tongguan Decoction＂ can improve the IL-1 and TNF-α in serum,and has better effects in 30 days than 15 days.
