Spinal Toll-like receptor 4 and neuropathic pain

摘要 Toll样受体4(Toll-likereceptor4,TLR4)是一类介导天然免疫的跨膜信号传递受体,在促进细胞因子的合成与释放、诱发炎性反应、抗病毒感染、调节免疫应答、介导全身免疫病理损伤等方面起重要作用。本文主要对TLR4的生物学特征、TLR4在神经病理性疼痛中的作用、TLR4参与神经病理性疼痛的可能机理和TLR4作为疼痛治疗的药物靶点等方面做一综述,以阐述脊髓TLR4在神经病理性疼痛发生发展中的作用,可能为临床神经病理性疼痛的治疗提供方向和思路。 Toll-like receptor 4 (TLR4) is a transmembrane signal transduction receptor for mediate innating immune response, which plays an important role in synthesis and release of cell factor, inflammatory reaction, antivirus infection, modulation of immune response and mediating systemic immune pathological damage. This review mainly summarized the biological features, the role in the neuropathic pain and the possible mechanism of TLR4 as a drug target in the treatment of pathological pain, especially the role of TLR4 in the progressing of neuropathic pain were illustrated. TLR4 may provide directions and ideas for clinical treatment of neuropathic pain.
