摘要
目的:探索艾叶中绿原酸的醇提工艺。方法:以乙醇为溶剂,在单因素实验的基础上,选取提取次数、提取温度、提取时间、料液比为自变量,绿原酸的得率为响应值,采用中心组合设计的方法,利用响应面分析方法,模拟得到二次多项式回归方程的预测模型。结果:艾叶中绿原酸醇提工艺的最佳条件为60%乙醇为提取溶剂、料液比1∶13、提取温度63℃、提取次数3次、提取时间60min,在此条件下,绿原酸的得率为0.820%。结论:通过对绿原酸提取工艺的研究,艾叶中绿原酸的提取率达94.04%。
Objective:In order to optimize ethanol extraction of chlorogenic acid in leaf of Artemisiae argyi.Method:A mathematical regression model for predicting extraction yield of chlorogenic acid as a function of ratio of independent variables,including extraction times,extraction temperature,extraction time and solvent/material ratio were established through fitting Box-Behnken central composite design experimental results.Interactive effects of the independent variables on extraction yield of chlorogenic acid were investigated by response surface methodology based on the analysis of single-factor experiments.Result:The optimal protocol for ethanol extracting chlorogenic acid from leaf of Artemisiae argyi was based on 60% ethanol as solvent,the ratio of material to solvent of 1:13(g·mL-1),extraction temperature of 63℃,extraction time of 60min and 3 times extraction,and extraction yield of chlorogenic acid could be up to 0.820%.Conclusion:The extraction ratio of chlorogenic acid in leaf of Artemisiae argyi was 94.04%.
