超声波协同纤维素酶对黄精多糖和皂苷的提取研究 被引量:25

摘要 以九华山黄精为原料,水为提取溶剂,采用超声波协同纤维素酶方法对其多糖和皂苷进行综合提取。通过单因素和正交实验研究了纤维素酶与底物质量比、超声时间、超声温度、浸提pH、超声功率对多糖和皂苷提取率的影响。结果表明:影响最显著的是纤维素酶与底物质量比,其次是浸提pH以及超声功率,超声时间和超声温度的影响相对较小。优化得出最佳提取工艺是纤维素酶与底物质量比1%,超声时间40min,超声温度55℃,浸提pH5.0,超声功率300W。此时的多糖和皂苷提取率分别是39.36%和11.69%。相比传统提取方法,大大提高了多糖和皂苷的提取率。 To use Jiuhuashan Polygonatum(Polygonatum cy-rtonema Hua)as raw material and water as solvent,synthetically of polysaccharides and sibiricoside was extracted from Polygonatum with cellulase enzyme under ultrasonic wave.The effect of the quality ratio of enzyme to substrate,ultrasonic time,ultrasonic temperature,pH,ultrasonic power on the extraction of polysaccharides and sibiricoside were investigated by single factor and orthogonal experiments.The results showed that the quality ratio of enzyme to substrate was the most significant factor affecting the synthetic extraction yield,the pH and ultrasonic power were the next place,and extraction temperature and extraction time effects were the least.The optimum extraction conditions were as follows:the quality ratio of enzyme to substrate 1%,ultrasonic time 40min,ultrasonic temperature 55℃,extraction pH5.0,ultrasonic power 300W.Under this condition,the yield of polysaccharides and sibiricoside were 39.36% and 11.69% respectively.The extraction rate of polysaccharides and sibiricoside had been greatly improved which compared to conventional extraction methods.
