摘要
目的建立RP-HPLC法测定奋乃静片的含量。方法采用Agilent C18色谱柱(5μm,4.6 mm×150 mm),柱温为30℃,以甲醇-磷酸盐缓冲液(取磷酸二氢钾1.36 g,加水溶解并稀释成1 000 ml,加三乙胺4 ml,用磷酸调pH至3.02)(70∶30)为流动相,流速为1 ml·min-1,检测波长为256 nm。结果奋乃静在8.048~40.24 mg·L-1的范围内线性关系良好(r=0.999 9,n=5),测得平均回收率为99.6%,RSD=1.09%(n=9)。结论该方法稳定可靠,重现性好,可有效控制奋乃静片的质量。
Objective To develop an HPLC method for the determination of the content of perphenazine tablets.Methods Agilent C18 column(5 μm,4.6 mm×150 mm)was used,the column temperature was 30℃,and the mobile phase was methanol-phosphate buffer solution(1.36 g potassium dihydrogen phosphate dissolved and diluted into 1 000 ml by water,added with triethylamine 4 ml,adjusted to pH 3.02 with phosphoric acid)(70∶ 30) at the flow rate of 1.0 ml·min-1.The UV detection wavelength was 256 nm.Results The linear range was from 8.048 to 40.24 mg·L-1(r=0.999 9,n=5).The average recovery was 99.6% with RSD of 1.09%(n=9).Conclusion This method is stable,reliable and reproducible,which can be used for the quality control of perphenazine tablets.
