摘要
目的:研究SLP-2基因在食管癌细胞系中的表达及其功能发生的具体分子机制。方法:运用West-ern-blotting检测SLP-2在TE12细胞系中的表达。利用PCR从TE12细胞的cDNA中扩增SLP-2的开放阅读框,定向连接到pEGFP-N3载体,并通过细胞免疫荧光实验检测SLP-2蛋白在TE7细胞内的表达。结果:SLP-2基因在TE12细胞中高表达,并成功构建pEGFP-N3-SLP-2荧光表达载体,SLP-2蛋白主要定位在细胞质内。结论:证明了SLP-2在食管癌细胞中高表达,其荧光表达载体的构建为进一步研究SLP-2在食管癌发生发展的分子机制奠定了基础。
Objective:To study the expression of SLP-2 gene and the molecular mechanism of its function in esophageal carcinoma.Methods:Western-blotting was used to detect the expression of SLP-2 in TE12 cell line.Full-length open-reading frame(ORF) of SLP-2 was amplified from cDNA of TE12 cells by PCR,and inserted into pEGFP-N3 plasmid.Cell immune fluorescence was used to detect expression of SLP-2 protein in TE7 cells.Results:SLP-2 was overexpressed in TE12 cell lines.pEGFP-N3-SLP-2 plasmid was constructed successfully,and GFP-SLP-2 mainly localized in cytoplasma.Conclusion:The overexpression of SLP-2 in esophageal carcinoma cells was proved;and the construction of pEGFP-N3-SLP-2 plasmid provided a useful clue for further research on the molecular mechanism of occurring and development of SLP-2 in esophageal carcinoma.
