Tyro 3受体酪氨酸激酶调控糖蛋白Ⅵ介导的血小板活化

摘要 目的研究Tyro 3受体酪氨酸激酶在糖蛋白Ⅵ(glycoprotein VI,GPVI)介导的血小板活化中的调控作用。方法以Tyro 3基因敲除小鼠的血小板为研究对象,应用GPVI的特异性刺激剂,在体外检测血小板的聚集和铺展能力,并应用流式细胞术分析血小板的颗粒释放及整合素αIIbβ3的活化情况。结果与野生型小鼠相比,Tyro 3基因敲除小鼠血小板的聚集和铺展能力减弱,颗粒释放标记物CD 62P的数量减少,整合素αIIbβ3的活化也受到明显抑制。结论 Tyro 3受体酪氨酸激酶在糖蛋白Ⅵ介导的血小板活化中具有重要的调控作用。 Objective To study the role of Tyro 3 tyrosine kinases receptor in glycoprotein Ⅵ(GPVI)-mediated platelet activation. Methods Platelets were isolated from wild type(WT) and Tyro 3 knockout(Tyro3-/-) mice,then we examined their responses to GPVI stimulation by measuring integrin αIIbβ3 activation,granule release,aggregation and spreading. Results Compared with WT platelets,Tyro 3-deficient platelets displayed a suppressed aggregation and spreading after GPVI stimulation,a reduced CD 62P expression and integrin αIIbβ3 activition also be observed. Conclusion Tyro 3 plays an important role in GPVI,mediated platelet activation.
