摘要
目的研究榄香烯对肺癌A549细胞真核起始因子4E(eIF4E)蛋白表达的影响及其作用机制。方法浓度为20μg/mL、40μg/mL、60μg/mL、80μg/mL、100μg/mL和120μg/mL的榄香烯、顺铂以及联合处理细胞后,Western blot法检查eIF4E蛋白表达,MTT法观察榄香烯对A549细胞的抑制作用,TUNEL法检测细胞凋亡。结果各实验组eIF4E蛋白表达水平在24 h和48 h均显著降低;MTT法、TUNEL法结果显示,榄香烯对A549细胞的抑制率及凋亡率呈浓度依赖性。结论榄香烯明显抑制肺癌A549细胞生长,其机制可能与下调eIF4E表达有关。
Objective To investigate the influence and mechanism of β-elemene on expression of eIF4E in non-small cell lung cancer A549 cells. Methods After cells treated with 20 μg/mL, 40 μg/mL, 60 μg(mL, 80 μg/mL, 100 μg/mL and 120 μg/mL β-elemene and cisplatin, Western blot was applied to detect protein expression of eIF4E; MTF was applied to observe the inhibition of 13-elemene to lung cancer cell in vitro; TUNEL was used to test the apoptosis. Results eIF4E protein expres- sion was significantly reduced in 24h and 48h. MTY and TUNEL showed that the inhibition rate and apoptotie rate of β-elemene on A549 cells indicated a dose-dependent property. Conclusion The proliferation of A549 cell was inhibited obviously by 13-elemene and the mechanism may be related with down-regulation of eIF4E expression.
