摘要 目的探索镍离子对L929细胞毒性作用规律。方法小鼠成纤维细胞L929在含不同镍离子浓度(0、200、400、800μmol/L)培养液中培养24、48、72h,倒置相差显微镜观察细胞形态变化,MTT法测定细胞相对增值率(RGR);培养24h,透射电镜观察细胞超微结构改变并拍照。结果 200~800μmol/L镍离子处理细胞不同时间点下各组间存在统计学差异(P<0.05)。倒置相差显微镜和透射电镜下,经不同浓度镍离子处理24 h后,细胞形态和亚结构出现不同程度的损害,出现细胞回缩、细胞核形态不规则、胞浆空泡性改变、线粒体肿胀等改变,且随着浓度增大,结构破坏更加明显。结论在镍离子刺激下,L929细胞增殖受到抑制,细胞形态及亚结构均有中毒样改变,在观察浓度及时间范围内,呈现浓度依赖和时间依赖趋势。 Objective To evaluate the cytotoxic effects of nickel ions on L929 cells and their cellular ultrastructural changes.Methods L929 cells were exposed to the culture mediums containing different concentrations of nickel ions(0、200、400、800 μmol/L)for 24,48 and 72h.MTT assay was used to assess the cytotoxicity of nickel ions.In addition,the relative cell proliferation rate(RGR) and the cytotoxicity grade were calculated.Cellular morphology and ultrastructural features were examined using transmission electron microscopy 24h after culture.Results The results of MTT assay showed that cell viability at different culture time points were higher than that in the control group,and the cytotoxicity grade was 3 or 4 when the concentrations of nickel ions were among 200-800 μmol/L.Ultrastructural alterations were observed including irregular shaped nuclei,lipid droplet formation and mitochondrial swelling,and those phenomena were dose dependent.Conclusions Cellular proliferation of L929 cell was obviously inhibited by nickel ions.Cellular morphology and ultrastructure of L929 became to be at toxic state,and that was dose-dependent and time-dependent.
