短暂全脑缺血的成年大鼠眼底荧光造影和病理形态改变

Fundus Fluorescein Angiography and Pathological Changes of Transient Cerebral Ischemia in Rats

摘要 目的建立短暂全脑缺血的成年大鼠视网膜微循环缺血模型。方法选取实验动物为Wistar雄性大鼠5只,采用Pulsinelli经典的大鼠四血管结扎模型,烧灼双侧椎动脉后,结扎双侧颈总动脉,缺血10 min后解除夹闭的缺血装置,恢复脑血流。对成活大鼠模型制成后,经麻醉、散瞳,行眼底荧光造影和病理形态检查,观察成年大鼠短暂全脑缺血的视网膜改变。结果全部5只短暂全脑缺血实验大鼠均显示视网膜微循环缺血,缺血大鼠眼底荧光造影:视网膜血管放射状充盈不完全;病理形态改变:视网膜血管不同程度充血,视网膜节细胞层少数细胞胞浆空泡变性。结论构建了视网膜微循环缺血实验模型,为今后临床药物的相关研究提供了理想的实验动物模型。 Objective To establish the model of retinal vasculature in rats of transient cerebral ischemia.Methods Five SD rats were used according to the procedure modified by Pulsinelli et al,four arteries in rats were prepared as follows:Bilateral vertebral artery was cauteried,bilateral carotid artery was ligated,and reperfused 10 min after cerebral ischemia.The rat model was examined with fundus fluorescein angiography and pathological observeration by anesthesia and expend pupil.Results All of five rats with transient cerebral ischemia showed retinal vasculature injure.Radial incomplete perfusion showed that retinal vasculation and cytoplasmic vacuolation was localized within ganglion cell layer of the retina.Conclusion The model of retinal vasculature in rats of transient cerebral ischemia was successfully constructed,it may be used in clinic drug research.
