齐墩果酸对兔耳增生性瘢痕的治疗作用

Effects of oleanolic acid on hypertrophic scarring in rabbit ear model

摘要 目的研究齐墩果酸(oleanolicacid)对兔耳增生性瘢痕的治疗作用。方法用打孔器产生创面建立兔耳增生性瘢痕模型。齐墩果酸(2.5%、5%、10%)外用给药,1次/d,连续22d。给药结束后切除瘢痕组织进行组织学检测并且测定I型和Ⅲ型胶原、基质金属蛋白酶.1(MMP-1)和生长转化因子βl(TGF-βl)的水平以及瘢痕增生指数(scarelevationindex,SEI)。结果用不同质量浓度的齐墩果酸给药22d后发现其对兔耳肥厚性瘢痕有显著的治疗作用,TGF-βl、I型和Ⅲ型胶原水平显著降低,MMP-1在瘢痕组织中显著增加,SEI明显降低。组织形态观察结果显示齐墩果酸对瘢痕有明显的改善作用。结论齐墩果酸对兔耳增生性瘢痕有治疗作用,可能成为治疗人类增生性瘢痕的活性药物。 Objective To investigate the effects of oleanolic acid, a naturally occurring triterpenoid, on hypertrophic scarfing in a rabbit ear model. Methods A rabbit ear model of hypertrophic scarfing was used, with wounds produced with a biopsy punch. oleanolic acid (2.5%, 5%, and 10%) was applied once daily to the scars for 22 d. On postoperative day 28, the scars were excised, and the tissue was used for histological examination and assays of the levels of collagens I and III, matrix metalloproteinase MMP-1, and transforming growth factor TGF-βl. The scar elevation index (SEI) was also determined. Results Treatment with different concentrations of oleanolic acid for 22 d significantly inhibited hypertrophic scarfing in rabbit ear tissue. Levels of TGF-βl, collagen I, and collagen III were significantly decreased and the level of MMP-1 significantly increased in the scar tissue. SEI was also significantly reduced. Histological findings showed significant amelioration of the scar tissue. Conclusion Oleanolic acid suppresses hypertrophic scarring in the rabbit ear model and may be an effective cure for human hypertrophic scarfing.
