摘要 为进一步阐明乙醇引起胃粘膜损伤的机理,本实验在离体培养的人胃上皮癌细胞株(BGC-823)建立了乙醇 诱导细胞凋亡的模型。从细胞形态、DNA梯状条带及流式细胞仪检测等方面,观察了不同剂量的乙醇作用不同时间 后BGC-823细胞凋亡的发生情况。结果表明,较低浓度的乙醇(1.5%~4%)能有效诱导细胞凋亡,且随剂量增大凋 亡发生时间缩短。较高浓度(大于4%)则引起细胞坏死。2%乙醇诱导时,6h即有调亡出现,12~18h DNA电泳呈 现梯状条带。提示细胞凋亡在乙醇引起胃粘膜损伤中可能有一定的作用。 To further elucidate the mechanism of ethanol-induced damage of gastric mucosa, a reliable model of ethanolinduced apoptosis in a gastric epithelial carcinoma cell line (BGC-823 cells) was established. The occurrence of apoptosis was observed under different doses of ethanol and different induction time. The criteria of judging apoptosis include cell morphology, typical DNA ladder and flow cytometry. The results showed that, at lower doses (l.5% -4%), ethanol could efficiently induce apoptosis and the latence of apoptosis shortened with the dose of ethanol increase. While higher (>4%) doses of ethanol caused necrosis. When ethanol was administrated at 2%, the morphological apoptosis appeared around 6h and DNA ladder appeared around 12-- 18h. The study suggests that apoptosis might play a role in ethanol-induced gastric mucosa damage.
