摘要
为简化PCR方法和减少标本用量,利用基因释放剂不提取DNA,比较了利用基因释放剂和不用基因 释放剂,对微量肝素抗凝血、微量结肠粘膜组织(置-20℃6个月)和含有PCR抑制剂的DNA标本直接进 行PCR扩增。结果,利用基因释放剂,可扩增出目的产物,不用基因释放剂,来扩增出目的产物。提示利用 基因释放剂直接进行PCR,简单快速,不用提取DNA,适于微量、含有PCR抑制物的DNA及陈旧组织标本 的检测。
In order to simplify PCR without extraction of DNA,and to detect the samples with PCR inhibitors,and to minimize the size of samples to a few microliters/micrograms,gene releaser was used. Heparinized blood samples of microliters,colonic mucosa tissues (stored in --20C over 6 months ) of micrograms and DNA samples with PCR inhibitors were checked with direct PCR with and without (as control ) gene releaser. PCR amplification product was positive with gene releaser,but negative without. Gene releaser is a very useful agent for the rapid and simplified direct PCR amplification without extraction of DNA,and suitable for detection of samples of microliters/micrograms and DNA with PCR inhibitors as well as tile long time stored tissues.
