急性肺损伤大鼠蛋白酶激活受体-1表达及其机制

摘要 目的探讨肺损伤后蛋白酶激活受体1(protease-activated receptor-1,PAR-1)的表达情况及其与炎症因子之间的关系。方法腹腔注射内毒素的方法复制大鼠肺损伤的模型,应用逆转录-聚合酶链反应(RT-PCR)技术检测PAR-1 mRNA表达,HE染色观察肺组织病理变化,酶联免疫吸附(enzyme-linked immunosorbent assay,ELISA)试剂盒检测炎性因子的改变。结果内毒素肺损伤时,PAR-1 mRNA随着脂多糖(lipopolysaccharide,LPS)干预时间的延长表达逐渐增加。肺内肿瘤坏死因子-α(tumor necro-sis factor-α,TNF-α)、金属基质蛋白酶-12(matrix metalloproteinase-12,MMP-12)升高与PAR-1的水平密切相关。结论 PAR-1的表达与TNF-α,MMP-12相关,PAR-1的抑制剂减少了肺组织MMP-12和TNF-α的释放,可能具有抗炎作用。 Objective To investigate the relationship between protease-activated receptor-1(PAR-1) and inflammatory cytokines in acute lung injury(ALI).Methods An ALI rat model was performed by intra-peritoneal injection of lipopolysaccharide(LPS),PAR-1 was analyzed by RT-PCR,enzymelinked immunosorbent assay(ELISA) assay for TNF-α,MMP-12.Results The PAR-1 expression was gradually up-regulated with the time after intraperitoneal injection of LPS.ELISA results showed that the level of TNF-α,MMP-12 increased,and was closely linked to the expression of PAR-1.Conclusion The expression of PAR-1 may be related to the production of TNF-α,MMP-12,and PAR-1 inhibitor reduces the production of TNF-α,MMP-12,it maybe play an anti-inflammatory role.
