槲皮素通过抑制己糖激酶的表达促进结肠癌SW480细胞凋亡 被引量:2

摘要 目的观察槲皮素对SW480细胞增殖和凋亡的影响并探讨其机制。方法采用流式细胞检测技术检测槲皮素诱导人结肠癌SW480细胞的凋亡能力;应用免疫细胞化学技术检测细胞中己糖激酶蛋白的表达。结果槲皮素以浓度依赖性的方式诱导结肠癌SW480细胞的凋亡并下调己糖激酶蛋白的表达。结论槲皮素通过抑制己糖激酶的表达来促进结肠癌SW480细胞凋亡。 Objective Investigate the apoptogenic activities of quercetin on SW480 cells.Methods Annexin V-FITC/PI stained fluorescence-activated cell sorter(FACS) was used to detcted the apotosis rate,Immunocytochemistry was used to examine cellular localisation and hexokinase protein levels.Results quercetin produced dose-dependent inhibition of SW480 cell growth and induced its apoptosis,the expressions of hexokinase proteins were down regulated by quercetin.Conclusion The quercetin induce SW480 cells apoptosis through downregulation of hexokinase
