摘要 【目的】探讨肝癌患者血清端粒酶逆转录酶(human telomerase reverse transcriptase,hTERT)mRNA表达水平及对肝癌的临床应用价值。【方法】设计人端粒酶逆转录酶基因引物和实时荧光PCR相对定量法反应体系,在ABI7500实时荧光PCR仪上检测45例肝癌患者,52例病毒性肝炎患者,29例肝硬化患者和58例健康人血清端粒酶逆转录酶mRNA表达水平并分析其差异。【结果】肝癌患者血清hTERT mRNA水平明显高于肝炎、肝硬化患者及健康人。肝癌患者血清hTERT mRNA相对表达量与肿瘤大小和分化等级及分期明显相关,与年龄、性别、以及肿瘤数量无明显关联。【结论】肝癌患者血清hTERT基因表达上调,其升高程度与肿瘤分化与进展相关,检测血清hTERT基因表达水平有助于肝癌的诊断和研究。 【Objective】 To investigate the expression level of hTERT in serum of hepatic cancer patients and its clinical significance.【Methods】 The mRNA of hTERT in serum were detected by real-time relative quantitative PCR method in 45 patients with hepatic cancer,52 patients with virus hepatitis,29 patients with hepatic cirrhosis and 58 healthy individuals.【Results】 The expression level of hTERT in serum of hepatic cancer group was significantly higher than other groups.The relative expression level of hTERT mRNA was demonstrated to be definitely correlated with tumor size,Edmodson grade and TNM stage,while not correlated with age,gender and number of tumors.【Conclusion】 The expression level of hTERT mRNA in serum was up-regulated significantly when the hepatic cancer occurred.It may provide a novel diagnostic and research tool for hepatic cancer.
