丹参素对子宫内膜癌KLE细胞体外抑制作用的实验研究

Investigation on the effects of suppressing KLE cell with danshensu in vitro

目的研究丹参素对子宫内膜癌KLE细胞生长的体外抑制作用。方法用等比稀释的7个浓度的丹参素及终浓度为60 ug/ml的顺铂作用于体外培养的KLE细胞,作用时间分别为12 h、24 h、48 h,分别应用荧光显微镜、酶联免疫检测仪、流式细胞仪分析丹参素对KLE细胞增殖、凋亡及细胞周期的影响。结果经丹参素作用后,子宫内膜癌KLE细胞生长和增殖明显被抑制,光镜下观察肿瘤细胞发生凋亡的细胞形态学改变;中性红检测丹参素组OD值明显低于阴性对照组,抑制率明显高于阴性对照组;24 h丹参素组细胞凋亡率明显高于其他对照组,细胞明显被阻止于G0-G1期,而S期、G2期细胞明显减少;以上差异均有统计学意义(P<0.05)。该差异具有时间及药物浓度依赖性。结论丹参素对体外培养的子宫内膜癌KLE细胞的生长有明显抑制作用,可通过促进其凋亡、影响细胞周期分布发挥其抗肿瘤作用。

Objective This study was designed to investigate the inhibition and induction of apoptos of danshensu on KLE cell in vitro.Methods KLE Cells were treated by different concentration of danshensu and OOP for 12,24 and 48 h.The morphous of KLE cells are measured by fluorescence microscope.toluylene red was used to measure the levels of the proliferation of KLE cells.The effects of salvia miltiorrhiza on cell cycle and apoptosis rate of KLE cell were observed by FCM.Results compared with negative control groups,danshensu could inhibit the growth 、 induce apoptosis and alter cell cycle in KLE cells in both time-and dose-dependent manners（P0.05）.Observed by fluorescence microscope,the morphous of KLE cells express apoptosised change. Conclusion Effect of danshensu on cultured endometrial carcinoma KLE cell growth inhibition,can promote their apoptosis,influence on the cell cycle distribution of its antitumor effect.
