不同染色方法对精子形态学评估的影响

The effect of different staining methods on sperm morphology assessment

目的:探讨Diff-Quik染色法及改良巴氏染色法对精子形态学评估的异同性及相关性。方法:对80例标本分别涂片两张,利用Diff-Quik染色法及改良巴氏染色法分别进行染色,并记录同一标本不同染色方法其正常精子百分率、头部缺陷精子百分率、颈部和中段缺陷精子百分率、尾部缺陷精子百分率、畸形精子形态指数(TZI)、精子畸形指数(SDI)等。结果:Diff-Quik染色法及改良巴氏染色法在精子形态评估中,正常形态率、各种类型的缺陷率(头部、颈部与中段、尾部)、畸形精子指数、精子畸形指数等均无统计学意义(P>0.05)。结论:Diff-Quik染色法及改良巴氏染色法在精子形态评估中具有较为一致的结果;改良巴氏染色法虽经典,但过于繁琐;在病人多且急需出具精子形态学检验报告的情况下,可以考虑使用Diff-Quik染色法作为精子形态评估染色方法。

Objective： To investigate the similarities and differences and relevance about sperm morphology assessment by Diff-Quik staining and modified Papanicolaou staining.Methods： Two slides were smeared in each case by Diff-Quik staining and modified Papanicolaou staining respectively for 80 cases,and the percentage of normal sperm,the percentages of sperm head defects,neck and middle defect and tail defects,the abnormal sperm morphology index（TZI）,the sperm deformity index（SDI） and so on were recorded.Results： There was no significant difference（P0.05）in sperm morphologyassessment between Diff-Quik staining and modified Papanicolaou staining.Conclusion：The results of Diff-Quik staining and modified Papanicolaou staining are consistent in sperm morphology assessment;the modified Papanicolaou staining is classical,but too cumbersome.Diff-Quik staining is the choice when pationts are in urgent need of sperm mcrphology inspection repart.
