摘要
建立了高效液相色谱法测定全血环孢霉素A 浓度的方法。全血样品经多步处理后以20 μL注入reosolve C18,5μ色谱柱,采用V(乙腈)∶V(水)= 80∶20 的流动相,在75℃柱温下,以1 m L/m in 流速洗脱,检测波长为208nm 。环孢霉素A 的回收率为99% ~104 % ,相对标准偏差< 3.42% (n= 5) ,日间< 8.19% (n= 4) 。最低检测浓度为25 μg/L。
A HPLC method for the assay of cyclosporine A in whole blood was estabtished.After multistep pretreatment ,20 μL sample was injected into reversed phase column (200 mm ×4.6 mm,5 μm,C 18 ) at 75℃,eluted with a mobile phase of acetonitrile/water(80∶20) at a flow rate of 1.0 mL/min and monitored at 208 nm.The analytical recovery was 99 %~104% and inter day and intra days RSD was below 3.4% and 8.2% respectively.The detection limit was 25 μg/L.The method provided excellent recovery and good precision,and was accurate.No interference was observed.
