摘要
为研究拟南芥核苷二磷酸激酶家族(AtNDPKs)5个成员之间的差异,对其在蛋白质结构、磷酸化位点、信号肽序列、功能区方面的差异进行了生物信息学的比较分析,并建立了系统进化树。结果显示,5个成员在等电点、吸光值和亲水性方面的差异明显。二级结构中,AtNDPK2的α螺旋比例与其他成员差异显著;在β转角和无规则卷曲方面,AtNDPK1a与AtNDPK3a表现出相反的情况。5个成员各含有1个NDK结构域,但分布位置不同。AtNDPK3a中酪氨酸磷酸化明显增多,AtNDPK1a的磷酸化位点与其他成员差异显著。AtNDPK1和AtNDPK2的信号肽裂解点位于第16和17位氨基酸之间,AtNDPK3和AtNDPK3a的位于第42和43位之间,而AtNDPK1a的位于第24和25位之间。AtNDPK1a的进化地位高于其他成员,AtNDPK3与AtNDPK3a的亲缘关系最近,而AtNDPK1与其他成员的亲缘关系较远。
In order to study the differences of the five members of nucleoside diphosphate kinase family in Arabidopsis thaliana(AtNDPKs),the bioinformatics analysis was performed on the protein structure,phosphoric sites,signal peptide sequence and functional domain,and the phylogenetic tree was constructed.The results indicated the five members of AtNDPKs have an obvious discrepancy in pI,extinction coefficients and hydropathicity.The ratio of alpha helix in AtNDPK2 is obviously different from others,and the ratios of beta turn and random coil in AtNDPK1a and AtNDPK3a are totally opposite.Each member of AtNDPKs has a NDK domain,but in different sites.The tyrosine phosphorylation of AtNDPK3a is obviously increased,and the phosphorylation sites of AtNDPK1a are obviously different from others.The cleavage sites of AtNDPK1 and AtNDPK2 are between pos 16 and 17,the cleavage sites of AtNDPK3 and AtNDPK3a are between pos 42 and 43,and that of AtNDPK1a located between pos 24 and 25.AtNDPK1a had the highest evolutionary status among members,and the genetic relationship between AtNDPK3 and AtNDPK3a is closest while AtNDPK1 is far from other members.
