摘要
以盾叶薯蓣顶芽为外植体,研究了升汞处理时间对无菌顶芽得率以及不同植物生长调节剂对顶芽增殖和生根的影响。结果表明:用升汞处理5min,盾叶薯蓣无菌苗得率最高,为84%。适宜的快繁培养基为MS+6-BA 2.0mg/L+NAA 0.2mg/L,培养30d时,顶芽繁殖系数为5.50,株高为6.08cm,茎粗壮,长势整齐。适宜的生根培养基为1/2MS+IBA 0.3mg/L,培养45d时,顶芽生根率可达97.23%。
To mass-scale product D.zingiberensis C.H.Wright,the apical buds of D.zingiberensis C.H.Wright was used as explants to study the effects of different sterilization times on the livability of the sterile apical buds,and the effects of plant growth regulators on rapid propagation and rooting of plantlets.The result showed that HgCl2 sterilization for 5 minutes was the optimal and the livability of the sterile apical buds was up to 84%.The rapid propagation medium wasMS+6-BA 2.0 mg/L+NAA 0.2 mg/L,where the propagation coefficient was up to 5.50 with 30 days,and the plantlet height was 6.08 cm with strong and regular plantlets.Compared with NAA and IBA,we got the optimal medium for rooting,i.e.1/2MS+IBA 0.3 mg/L,where the rooting rate was up to 97.23% within 45 days.
