摘要
目的探讨神经生长因子(nerve growth factor,NGF)基因转染对牙周膜细胞(periodontal ligam entcells,PDLC)成骨性的影响。方法运用基因转染技术将NGF基因通过质粒pcDNA3.1-NGF转入PDLC中,利用免疫组化及免疫印记法鉴定转染细胞的表达并观察对其成骨性的影响。结果免疫细胞化学证实,牙周膜细胞有NGF蛋白表达。转染NGF后的PDLC的OC含量较未转染NGF组明显增高(P<0.01)。结论转染NGF后的牙周膜成纤维细胞成骨性明显增强,为使牙周膜细胞成为牙周组织再生的种子细胞奠定基础。
Objective To explore the osteogenic effect of nerve growth factor gene transfection on periodontal ligament cells.Methods NGF gene was transfected into human periodontal ligament cells through plasmid pcDNA3.1-NGF by using gene transfection technique.Immunohistochemical method and Western blotting were used to identify the expression of transfected cells and to observe osteogenic effects.Results The results of immunocytochemistry showed that the NGF protein was expressed in periodontal ligament cells.The OC content in the PDLC transfected by NGF increased obviously compared with the group that were not transfected.(P〈0.01).Conclusions The osteogenic ability of human periodontal ligament fibroblasts increased obviously after being transfected by NGF,so as to set a basis to make PDLC become periodontal regenerated seeding cells.
