摘要 建立高效液相色谱法分离、蒸发光散射检测器(HPLC-ELSD)测定酸羊奶中水苏糖含量的方法.采用Hypersil NH2(250mm×4.6mm,5μm)色谱柱;流动相为甲醇∶水(83∶17),流速1mL/min,柱温为室温;蒸发光散射检测器(ELSD)参数,漂移管温度85℃,氮气压力35Psi,增益10.结果表明,在上述条件下测得水苏糖在2.504~12.52μg范围内线性关系良好,相关系数为0.999 8.本方法操作简便、稳定,结果准确、灵敏度高、重现性好. To establish a HPLC-ELSD method for determining the stachyose in goat yogurt. Stachyose was successfully separated on a Hypersil NH2 (250 mm ×4.6mm,5μm)column at ambient temperature, the mobile phase composition was Methanol:water (83 : 17). The flow rate was 1. 0 mL/min. The parameters of ELSD were set as follows: Drift tube temper- ature was 85℃, Gas pressure was 35 Psi ,Gain was 10. This method fulfilled all the standard requirements of linearity, accuracy and precision. The linear range was 2. 504-12.52 /2g (r=0. 999 8). This method was simple, stable, accurate, sensitive, and reproducible.
