大黄与虫对药含药血清对胰岛素样生长因子-I和表皮生长因子促进人子宫肌瘤细胞增殖的影响

Effect of Dahuang and Zhechong medicated serum on proliferation of primary cultured human leiomyoma cells in vitro promoted by IGF-1 and EGF

目的:观察大黄虫对药不同剂量含药血清对胰岛素样生长因子-I(IGF-I)和表皮生长因子(EGF)促进原代培养人子宫肌瘤细胞增殖的影响。方法:原代培养并建立人子宫平滑肌瘤细胞系,采用中药血清药理学方法处理细胞,以MTT比色法检测不同剂量含药血清作用于经IGF-I、EGF处理的子宫肌瘤细胞12、24、48、72h的光密度值,计算细胞存活率。结果:大黄虫对药含药血清可抑制IGF-I、EGF对子宫肌瘤细胞的促增殖作用。结论:大黄虫对药含药血清通过抑制IGF-I、EGF对子宫肌瘤细胞的增殖作用,发挥其抑制肌瘤生长的作用,这可能是该对药治疗子宫肌瘤的机理之一。

Objective： To observe the effect of Dahuang and Zhechong medicated serum on proliferation of primary cultured human leiomyoma cells in vitro promoted by IGF-1 and EGF. Methods： Human leiomyoma cells were primary cultured in vitro. Treated the cells with IGF-1 and EGF, then Chinese herbs serum pharmacology method were used acting on the cells. The MTT assay was adopted to investigate the survival rate of 12h, 24h, 48h, 72h after the cells were treated. Results： Serum containing Dahuang and Zhechong showed an antagonistic effect on proliferation of uterine leiomyoma cells promoted by IGF-1 and EGF. Conclusion： Dahuang and Zhechong medicated serum has inhibitory effect on proliferation of hysteromyoma cells promoted by IGF-1 and EGF, it may be one of mechanism that Dahuang and Zhechong treating uterine leiomyomas.
