摘要
目的:建立夏枯草药材高效液相色谱(HPLC)特征图谱分析方法,为科学评价夏枯草药材质量提供参考。方法:采用Diamonsil C18色谱柱(4.6mm×250mm,5μm),甲醇-0.1%的磷酸水溶液梯度洗脱,流速1.0mL/min,检测波长210nm,柱温:30℃。所得不同样品的特征图谱用相似度评价软件进行相似度分析,用SPSS进行聚类分析。结果:建立了夏枯草的特征图谱,分析了10个产地夏枯草药材的HPLC特征图谱,经相似度分析,确定了29个色谱峰为夏枯草药材特征峰,指认了其中的8个共有峰。结论:该方法简便、准确、重现性好,研究结果初步说明了不同产地夏枯草药材的差异与相似性,为夏枯草的质量评价和质量控制提供了参考。
Objective: To establish the method of fingerprint analysis on Prunella vulgaris by HPLC, which could be used for quality evaluation of herbs Prunella vulgaris. Methods: Column of Diamonsil C18 (4.6mm×250mm, 5μm) was adopted with a gradient elution composed of methanol- 0.1% phosphoric acid solution as a mobile phase at flow rate of 1.0mL/min and detection wavelength was 210nm, and column temperature was 30℃. Evaluated the similarity of the 10 samples by the software of ‘traditional medicine fingerprint similarity evaluation', and Cluster analysis has been carded out using SPSS. Results: Established the method of fingerprint analysis on Prunella vulgaris by HPLC, twenty-nine characteristic peaks were established in the fingerprint of 10 samples, among them 8 peaks were identified. Conclusion: The method is simple and accurate with good reproducibility. It can be used for the quality control of Prunella vulgaris.
