滨蒿总黄酮提取物的抗流感病毒作用 被引量:17

Study on the effect of total flavonoids extracted from Artemisa scoparia on anti-influenzavirus

摘要 目的:观察滨蒿总黄酮提取物(滨蒿总黄酮)在体内外的抗流感病毒作用。方法:体外抗病毒实验观察样品对狗肾传代细胞(MDCK)的毒性,观察病毒致细胞病变(CPE)法测定样品在体外抑制甲、乙型流感病毒的半数抑制浓度(IC50),并计算选择指数(SI)。体内抗病毒实验用流感病毒鼠肺适应株(PR8)滴鼻感染小鼠建立流感病毒性肺炎模型,以死亡保护率、生命延长率和肺指数等为指标,评价样品的保护作用。结果:滨蒿总黄酮对甲、乙型流感病毒致细胞病变的IC50分别为74.6μg/mL、98.5μg/mL,SI分别为16.8、12.7;滨蒿总黄酮以200、400mg/kg剂量灌胃给药对小鼠流感病毒性肺炎模型有明显的保护作用,其死亡保护率分别为36.4%、54.5%;生命延长率分别为29.0%、38.2%;滨蒿总黄酮400mg/kg剂量对流感病毒性肺炎小鼠肺指数有明显的抑制作用,抑制率为25.9%。结论:滨蒿总黄酮在体内外均有明显的抗病毒作用。 Objective: To investigate the active of the total flavonoids from Artemisia scoparia on anti-influenza virus both in vivo and in vitro. Methods: In vitro the cell culture technique was used in MDCK cells to investigate the toxic concentration of drug. The IC50 of drugs was determinated and the SI was calculated in vitro inhibition of A and B influenza viruses by CPE. In vivo the mice were infected with influenza virus PR8 from nose. The death rate, mean survival days and lung index of infected mice were observed to investigate the protective action of the sample on anti-influenza virus in vivo. Results: The ICs0 of the sample to cytopathic CPE caused by A and B influenza viruses were 74.61ag/mL, 98.51ag/mL, respectively, and the SI were 16.8, 12.7, respectively. At the dose of 0.4g/kg and 0.2g/kg, it had the remarkably protective action to flu viral pneumonia model in mice, its protective rate were 36.4%, 54.5%, respectively, and the extending lifetime rate were 29.0%, 38.2%, respectively. At the dose of 400mg/kg, it had the significantly inhibited action to the pulmonary index with the inhibition rate was 25.9%. Conclusion: The total flavonoids from Artemisia scoparia had better effect against the test virus both in vivo and in vitro.
