摘要
目的研究Ent-11α-hydroxy-15-oxo-kaur-16-en-19-oic-acid(以下简称5F)对人高转移巨细胞肺癌PGCL3细胞毒性、侵袭能力及PTEN、基质金属蛋白酶-9(MMP-9)表达的影响。方法 MTT法检测5F对PGCL3细胞增殖抑制的影响;用Transwell小室法检测5F对PGCL3细胞侵袭能力和趋化性运动能力的影响;RT-PCR和Western blot方法检测5F对PGCL3细胞侵袭转移相关基因PTEN、MMP-9表达水平的影响。结果 5F对PGCL3细胞的增殖、侵袭及运动能力均有明显的抑制作用(均P<0.05)。5F作用于人PGCL3细胞24h,能明显上调PTEN mRNA和蛋白的表达,下调MMP-9mRNA和蛋白的表达。结论 5F能抑制PGCL3细胞的增殖、侵袭和迁移能力,其机制可能是通过上调PTEN和下调MMP-9表达水平而实现的。
Objective To investigate effect of envlla-hydroxy-15-oxo-kaur-16-en-19 oic-acid(5F)on the cell growth inhibition and invasion, migration and the expression of PTEN and MMP-9 in the human highly metastatic huge cell lung cancer PGCL3 cells in vitro. Methods MTT assay was used to detect the growth inhibition by 5F in PGCL3 cells;Transwell chamber assay was performed to detect the effect of 5F on invasion and migratory capacity of PGCL2 cells;The expression level of PTEN and MMP-9 was examined by using reverse transcription PCR(RT-PCR)and Western blot. Results 5F could inhibit the abilities of proliferation,migration and invasion of PGCL3 cells in vitro (P〈0.05). After PGCL3 ceils were treated with 25 and 50 gmol/L 5F for 24 h,the expression of PTEN was up-regulated and that of MMP-9 was down-regulated. Conclusion 5F can inhibit the migration and invasion of PGCL3 cells in vitro probably be up-regulating PTEN expression and down-regulating MMP- 9 expression. 5F might be a potential drug to inhibit metastasis of lung cancer.
