摘要
目的 研究山莨菪碱 (Ani)对家兔心室肌细胞L 型钙通道的电生理作用。方法 采用全细胞膜片钳 (Wholecellpatchclamp)技术 ,当维持电位为 - 40mV ,刺激频率为0 5Hz ,钳制时间为 2 5 0ms,步进电压为 10mV ,去极到 6 0mV ,观察不同浓度的Ani对L 型钙通道电流 (L ICa)的影响。结果 0 1mmol·L-1Ani对L ICa无影响 (P >0 0 5 ) ;0 2、0 4和 0 8mmol·L-1Ani对L ICa的抑制为 36 3% ,5 1 1%和 72 8% (P均 <0 0 1) ,且浓度愈大 ,其抑制作用愈强 ,但都不使I U曲线的峰值发生偏移。结论 Ani能浓度依赖性阻滞L ICa,具有钙拮抗作用。
AIM To study the cardiac L type calcium channel electrophysiologic effects of anisodamine (Ani) on rabbit ventricular myocardium METHODS When the holding potential was -40 mV, cells were depolarized to 60 mV for 250 ms with steps of 10 mV at frequency of 0 5 Hz by whole cell patch clamp technique RESULTS 0 1 mmol·L -1 Ani did not affect the L type calcium current (L I Ca ) (P>0 05) 0 2, 0 4 and 0 8 mmol·L -1 Ani inhibited L I Ca by 36 3%, 51 5% and 72 8%(P all <0 01), respectly, without shifting the current voltage (I U) curve for the peak of L I Ca CONCLUSION The Ani can inhibit L I Ca in a concentration dependently manner and has calcium antagonistic effect
