低氧高碳酸性离体膈肌疲劳模型的建立 被引量:3

摘要 目的 建立低氧高碳酸性大鼠离体膈肌疲劳模型。方法 用低氧高二氧化碳混合气 (2 1%O2 ,12 %CO2 ,6 7%N2 )造成膈神经 膈肌标本疲劳 ,用计算机输出电刺激并记录张力的变化。结果 混合气降低了 5~ 12 0Hz所有频率下的张力 ,且使峰颤搐张力、维持张力、肌刺张力均下降 ,预1999 0 6 0 3收稿 ,1999 0 8 12修回作者简介 :徐旋里 ,女 ,2 5岁 ,硕士研究生 ;杨秋火 ,男 ,62岁 ,教授 ,浙江省生理科学会理事长。研究方向 :呼吸生理、药理先加入氨茶碱 6× 10 -4 mol·L-1及 1× 10 -3 mol·L-1明显提高了所有频率下的张力 ,同时提高了峰颤搐张力、维持张力及肌刺张力。结论 通低氧高二氧化碳混合气 4min能够造成低氧高碳酸性大鼠离体膈肌疲劳模型。 AIM To set up a fatigued diaphragm model in vitro due to hypoxic、 hypercapnic acidosis. METHODS The fatigued diaphragm model was set up with 21% O 2,12% CO 2,67% N 2 gas mixture, and computer was used to put out electric stimulation and to record diaphragmatic contractile force simultaneously. RESULTS Gas mixture decreased force at each frequency(5~120 Hz),and decreased peak twitch tension、maintaining tension and tension by muscle stimulation significantly.Aminophylline(6×10 -4 mol·L -1 ,1×10 -3 mol·L -1 )restored force at all frequencies of stimulation,and improved peak twitch tension、maintaining tension、and tension by muscle stimulation. CONCLUSION A model of rat diaphragmatic fatigue in vitro induced by hypoxic、hypercapnic acidosis can be established by aerating with 4 min gas mixture(21%O 2、12%CO 2、67%N 2).
