摘要
通过PCR扩增获得了3种中国黄渤海海域的鳚亚目鱼类的线粒体ITS-1基因,并以条斑马鲛为外群,生成供系统发育分析的序列矩阵,利用MEGA 4.0软件分析序列的碱基组成、差异百分比、转换/颠换值等,应用最大简约法(MP)和邻接法(NJ)构建系统发育树。结果表明:在鳚亚目鱼类的ITS-1片段生成的序列矩阵中发现有碱基的插入缺失现象,共有49 bp变异位点,转换/颠换值为1.0;3种鱼ITS-1长度为375~562 bp,其中方氏云鳚的最长,为532~562 bp,六线鳚的最短,为375 bp;锦鳚科的方氏云鳚和云鳚聚为一支;方氏云鳚和云鳚种间遗传距离只有0.02,亲缘关系最近。
In this study,the internal transcribed spacer 1(ITS-1)partial sequences of 3 species from Blennioidei in China yellow and bohai seas were amplified using PCR technique,Scomberomorus regalis was designed as outgroup.Nucleotide composition frequencies,Kimura-2 parameter genetic distance and transition/transversion ratios were analyzed with MEGA 4.0 software.Molecular phylogenetic trees were constructed by maximum parsimony(MP)and neighborjoining(NJ)methods also with MEGA 4.0.The results were as follows:There were insertions and deletions in the aligned base pairs,49 bp variable sites were found in the analysis matrix.,the average ts/tv ratio was 1.0;The length of ITS1 was from 375 bp to 562 bp,with the Enedrias fangi′s the longest,and the Ernogrammus hexagrammus the shortest.Enedrias nebulosus and Enedrias fangi were placed in a same clade with support of a high bootstrap value.Enedrias fangi and Enedrias nebulosus had a close relationship,as the genetic distance between the two species of ITS-1 was only 0.02.
