摘要
目的研究运动大鼠骨骼肌细胞线粒体膜电位的变化与细胞凋亡的关系。方法用流式细胞仪 (flowcytometry ,FCM)检测肌细胞线粒体膜电位、DNA倍体分析及脱氧核糖核苷酸末端转移酶介导的缺口末端标记技术 (terminaldeoxynucleotidyltransferasemediatedd UTPnickendlabeling ,TUNEL)。 结果线粒体膜电位在游泳运动后第 1周降低 ,第 2周后开始明显增高 ,第 3周略有降低 ,运动后各组的DNA均出现亚“G1”峰即凋亡峰。TUNEL显示 ,运动后第 1周凋亡比例最高 ,第 2周后比例降低。结论运动引起线粒体膜电位的改变 ,该方法快速、特异性强。
ObjectiveTo investigate the relationship between mitochondrial membrane potential change, and apoptosis of muscular cells in rats after exercise.MethodsMitochondrial membrane potential in the muscular cell was determined with analysis of DNA double by the technique of flow cytometry,and terminal deoxynucleotidyl transferase d UTP nick end labelling(TUNEL) test was conducted. ResultsThe mitochondrial membrane potential decreased in one week, then increased markedly in two weeks, and it become much lower compared with control by three weeks after exercise. DNA double analysis demonstrated that sub G 1 namely apoptosis peak appeared at different level in three groups after exercise. TUNEL showed that the apoptosis rats augmented after exercise compared with control rats, and the rate reached the highest value after one week.ConclusionExercise may cause mitochondrial membrane potential change and induce muscular cell apoptosis.The procedures of these measurements are performed quickly with high specificity and sensitivity.
