两种异位种植结肠癌肝转移动物模型的比较 被引量:8

摘要 目的建立并对比两种异位种植结肠癌肝转移小鼠模型。方法以1×106个/mL小鼠结肠癌细胞株(CT26)0.2mL对BALB/c小鼠分别行肝门静脉注射法,脾脏种植切除脾脏法构建结肠癌异位种植肝转移动物模型。术后待小鼠自然死亡,比较两种动物模型的肝转移率和成瘤效果、肺转移率以及小鼠生存期的差异。结果肝门静脉注射法与脾脏种植切除脾脏法相比,前者在肝转移率、肝脏成瘤效果和肺转移率方面高于后者,后者在小鼠生存时间和操作难易程度方面优于前者。结论本研究建立并对比了两种具有高转移率的结肠癌肝转移小鼠模型,肝门静脉法在肝转移率和肝脏成瘤效果方面更具优势,适合对肝转移率和取材要求较高的实验研究;而脾脏种植切除脾脏法则更适合验证周期较长的药物实验。 Objective To establish and compare two kinds of ectopic implantation of colon cancer liver metastasis animal model.Methods We establish two kinds BALB/C mice colon cancer liver metastasis models of hepatic portal vein injection and spleen planted add splenectomy by using 1×106 /mL murine colon carcinoma cell lines(CT26).After surgery and until the mice died of natural causes,we compare the two animal models of liver metastasis rate and tumorigenic effects,the difference of the incidence of lung metastasis,and survival time of mice.Results Comparing hepatic portal vein injection model with removal of the spleen after spleen planting method,the former were higher in the rate of liver metastases,the effect of liver tumor formation and lung metastasis than the latter,the latter were higher in terms of survival time and operating ease.Conclusion This study established and compared the two high colon cancer liver metastasis mouse model,the hepatic portal vein injection model take advantage in the effect of liver metastasis rate and liver tumor formation,which is suitable for higher liver metastasis rates and material experimental study;removal of the spleen after spleen planting is more suitable to verify a longer period of drug experimentation.
