摘要
采用平板培养法对毛栓孔菌(Trametes hirsuta)菌丝体营养条件进行优化研究,通过单因素和正交试验的方法筛选出菌丝体生长的最佳培养基。试验结果表明:毛栓孔菌菌丝体生长的最适碳源为蔗糖,最佳氮源为酵母膏,最佳的碳氮比是101,最佳的无机盐是K2HPO4和Na2SO4,在基础培养基中添加复合维生素对菌丝体的生长影响显著,通过正交试验获得最佳的培养基:蔗糖4%,酵母膏1.3%,磷酸氢二钾0.05%,硫酸钠0.2%。
In this study,the cultural medium for the growth of mycelium of Trametes hirsute was examined by using the method of plate culture From the results of one-factor-at-a-time and orthogonal method,the best carbon and nitrogen source for the growth of mycelium of Trametes hirsute were cane sugar and yeast extract respectively,while the optimal of carbon-nitrogen ration was 101.Among the mineral salts,K2HPO4 and Na2SO4 supported the maximal mycelium growth.Adding vitamin to basic medium had significant influence on the growth of mycelium.Based on the result of orthogonal method,the optimized medium contained cane sugar 4%,yeast anointed 0.13%,K2HPO40.05%,AND NA2SO40.2%.
